Hi,
I am trying to create a very large contact sheet with 512 columns of images. Is it possible to adjust the contact sheet script to allow me to have 512 columns rather than the maximum 100?? Or is there any other ways this could be achieved?
Any help would be much appreciated!

If you're talking about the Contact Sheet that is built into Photoshop, I don't believe that is a script, so I doubt that can be changed. If you have a separate Contact Sheet script that someone has written, then you should be able to do it, depending on the complexity of the script.
What OS are you using? There might be better tools for this than Photoshop's Contact Sheet. If you're on a Mac, then GraphicConverter should allow you to enter that many columns. Plus it has a lot more features and options available for making contact sheets.

Sorry, although I use PCs as well as macs, whenever I need to print a contact sheet, I use GraphicConverter on the Mac, so I'm not sure what programs might be available to do this on a PC. I believe someone did write a Javascript contact sheet script, but I wasn't able to find it through a quick search. Perhaps someone else might be able to help verify this.

ContactSheetX (aka CSX) is a PS script at ps-scripts.sourceforge.net. The number of rows and columns are hardwired at one place in the script to 30 each.
It would be easy enough to change that to 512 or whatever.

Xbytor's script is the way to go in Photohop.
