How we can make two channels with seperate colours ?

Figure shows random paths (Circle type). One path is drawn dividing circle type path to two unequal one. How to make to different channels filling different colours.

Why Channels and not Layers?

If the Question concerns the Paths:

If you complete the intersecting line to an area covering one half of the path you can use Path Operations to exclude or intersect it.

Spot colors (and the spot channels that house them) are usually used when a special color is needed to enhance a four-color rendition (such as adding peach and warm gray to a flesh tone: a 6-color job), to print in a metallic color or to print a solid PMS color rather than a CMYK "close match" tint mixture. That does not appear to be something you want here. Rather, as c.pfaffenbichler suggested, you should be creating your desired result using Layers. If, in fact, you want spot channels, please repeat the request for creating them and I will add instructions.

Any thing is ok layers and channels. Was finding difficulties with layers so was trying with channels.

If I understand your intentions correctly it would seem fairly easily done with Layers.

I do not understand you question.  Alpha channels are like a selection your selection area not about color about . These are a grayscale map of your selection White area is shown, black area is hidden and gray area partly transparent when used as a mask.  You can have up to 53 Alpha Channels.  Additionally you can had Layer mask and smart filter mask which are like alpha channels associated with a layer or group they only show in the channel Palette when the layer or group they are attached  to is the current Photoshop target. You can load an Alpha Channel as a selection and fill the mapped area on a layer with any color you want.  You just need your two alpha channel selections to color two area.

But that is intended for actually printing with Spot Colors.

That well may be I never been involved with commercial CMYK printing there are many feature in Photoshop I have little knowldge about. I know that Spot Colors exist but not how they are used or done in Photoshop.

If this is regarding printing plates… Either keep it in RGB/CMYK with the “extra” composite and colour channels or change to Multichannel mode, deleting unwanted channels. Then the remaining channels would need to be made a spot colour channel, and most likely inverted so that the background was white and the printed areas black or shades of gray.
