Are you able to select a 'Location' for your file to export to at all? Can you try toggling any of the 'Destinations' and 'Publish' buttons on the left on and off, and then go back to the main Media File export to see if you can select or change the file export location?

As a potential workaround, are you able to drag your sequence from Premiere into Media Encoder and export from there?
