Unfortunately Illustrator needs a printer, although that shouldn't stop it from starting. In case you don't have a printer, you might want to install a PDF printer. When Illustrator launches then in the startup screen it reads what it does, like "Loading fonts", Loading plugin XY" ... this gives hints. Unformtunately Illustrator doesn't behave too well with font managers. FontBook should be fine. FontBook also allows you to check your fonts for corruption. Also Wacom drivers are a main cause of trouble. Check if there's a new one available. Also try and uninstall it and detach the tablet, just to check if it's the issue. User accounts (on your system) tend to get corrupt. To find out if this happens you can set up a new one, launch into that and then see if Illustrator works. This will also create a new set of preferences and of course there are no user fonts installed.
