To lock or unlock all composition markers on a composition, right-click (Windows) or Control-click (Mac OS) a marker on the composition marker.

That's a funky template. The first red flag is that the control layer, with all the sliders and color controls, is on an adjustment layer instead of a Null. The second one is that sliders are used for everything when Menu, checkbox, and point controllers could simplify the UI and make setting things up a lot easier.

Scripts cannot lock or unlock markers. You generally can only create or modify properties that have keyframes or will receive values.
