Probably you are using the Cinema 4D renderer. Enter into your composition Settings and check what renderer are you using in the 3D Renderer Tab. If so change to Classic 3D and check if your layers are visible.

The biggest problems that I see are the use of the C4D rendering engine and the Comp size. Unless you are an expert in video formats and compression you should be using standard, not custom, comp presets. The C4D rendering engine disables a lot of features and has no real advantage for this kind of project.

A minor problem that I see is that the comp resolution is set to Full and the Magnification Factor is set to 25%. Setting the Comp resolution to Auto will speed up your workflow and give you previews that respond better to the magnification factor for the comp panel.

If Layer 1, Tammy, disappears when you make it 3D then layer 2, Background, is probably in front of layer 1. When you have problems with a composition, the very first thing to do is to select the layers that are giving you problems, press the U key to show everything you have modified on the layers and start resetting things or turning off effects until you isolate the problem. If you are having problems with 3D layers then enable multiple comp views (at least 2) and check the position of your 3D layers. The only view that will render is the Active Camera view, so make sure you are using that view to set up your project.
