Help w AE and Unity..Transparency

Hi good morning everyone,

Recently I've taken up a task to help a friend with a game he's building. I decided to make an animation of a cube with a ring going around it a couple times- it's 3d.

He would like to be transparent so he can place in unity with a movie texture anywhere on the game.

He wants the file size to be as small as it could be. The animation is done but I have spent two days trying to research and come up with how to export it with transparency with smaller file size.  I've used Quicktime with JPEG2000, PNG, Animation etc. Though are not supported on windows media player. He needs it to be supported if he wants to put it in his game.  Unity can't import a file that relies on quicktime only.

If I've overlooked anything please help out,
