Is there any way to use expressions to get a list of all system fonts?

No. Expressions cannot access operating system details directly.

As Rick says you can't use Expressions to do that, but what is it you're trying to achieve?  What's the end result?  With After Effects, there's generally a way to get there.

This page on Microsoft's website lists the fonts included.  If nothing else, you should be able to copy and paste the names into a Text layer:

Trying to create a MOGRT with a fonts selection dropdown.  However I've just discovered that source text in the EG panel has the option to change font and faux options which is perfect 🙂
