I'm using Adobe After Effects CC v16.1.2 on a Mac running OS High Sierra 10.13.6. I've created an animated 3D "block" (square shape with extrusion and bevel) and can't remove the transparency of the object. I've turned off Draft 3D and the blend mode is normal. Any suggestions for a newbie. Thanks for any assistance.

please, show a screenshot with Material section expanded. It sounds like you have some transparency applied to the object.

Your screenshot is not any help in diagnosing the problem. We need to see the fill comp with the modified properties of the layer or layers that are giving you problems. The most likely problem - Many modes and effects are turned off if you choose the C4D rendering engine or Ray-traced rendering. You have to read the warnings. There are workarounds, but without a complete description of your workflow and/or a detailed screenshot we have no idea what kind of user error you have made.
Select the layer or layers, press "uu" to reveal all modified properties, PrintScreen and Paste or Drag to the forum.
EDIT: I see screenshots of part of your project that you posted while I was writing my reply. What are the comp settings? I also don't see the extrusion settings. Press UU so we can see what you have modified.

OK, thanks, and how does it look in Composition window?
Without that it's quite difficult to say something.
Meanwhile, try to change and check settings of 3D Renderer (Composition Settings > 3D Renderer). It seems that now you use Cinema 4D renderer, it does not allow to set transparency. Try Ray-Traced 3D and make sure that Transparency is 0%

What do you mean when you say you "can't remove transparency?"
Here is a square shape layer turned into a Cube with the C4D rendering engine and the opacity reduced to 59%.
Here are the limitations of C4D rendering:
I'm not sure what you are trying to do that you can't do.

but your rendering result is not transparent. If you mean that M letter is visible through the box, it's not so. M letter is, as far as I can see, the separate layer, you have to check their interaction.

I have the bevelled initial mirroring the cube's movement/positioning. With no transparency applied to the cube's materials/visibility, why am i able to see the initial?

because now it is not behind the box. It's in fron of the box. If the box was transparent, we could see its color in non-opaque mode. But now initial shields the box. Check their interaction. Maybe the adjusting layer causes some problem.
